Putative Ca2+ P-type ATPase, TMEM94, of 1356 aas and 10 TMSs in the usual 2 + 2 + 6 TMS arrangement.  This protein is very distantly related to all other members of the 3.A.3.2 family within the P-type ATPase superfamily, and therefore may have a different or unique function (Zhang et al. 2018). It may functioin with NPC2 of 123 aas and 1 N-terminal TMS. ERMA (TMEM94) transports Mg2+ uptake in the endoplasmic reticulum (Vishnu et al. 2024).  The ER is a major Mg2+ compartment refilled by TMEM94.  ERMA uniquely combines a P-type ATPase domain and a GMN motif for ERMg2+ uptake. A tyrosine residue is crucial for Mg2+ binding, and activity is a mechanism conserved in both prokaryotic (MgtB and MgtA) and eukaryotic Mg2+ ATPases. Cardiac dysfunction by haploinsufficiency, abnormal Ca2+ cycling in mouse Erma+/- cardiomyocytes, and ERMA mRNA silencing in human iPSC-cardiomyocytes collectively define ERMA as an essential component of ERMg2+ uptake in eukaryotes (Vishnu et al. 2024).
