Serine/threonine-protein kinase component of the WNK1-SPAK/OSR1 kinase cascade, which acts as a key regulator of blood pressure and regulatory volume increase by promoting ion influx (Boyd-Shiwarski et al. 2022).  WNK1-dependent water influx is required for CD4+ T cell activation and T cell-dependent antibody responses (Biggs O'May et al. 2025).  This protein contains 2382 aas and several potential TMSs.  Phosphorylation of Na-K-Cl cotransporters SLC12A2/NKCC1 and SLC12A2/NKCC1 promotes their activation and ion influx; simultaneously, phosphorylation of K-Cl cotransporters SLC12A5/KCC2 and SLC12A6/KCC3 inhibits their activity, blocking ion efflux (Rinehart et al. 2009, Richardson et al. 2011).  It also acts as a negative regulator of autophagy (Gallolu Kankanamalage et al. 2016).
